Automated sample preparation methods for mixing and centrifugation

This poster describes two applications that benefited from centrifugation in the sample extraction and clean-up, using Anatune’s solvent safe CF200 centrifuge.  The first was a methanolic extraction of soils for the analysis of BTEX followed by GC/MS analysis, this method showed good linearity for Benzene, ethyl benzene, toluene, and the xylenes.  The other was a pharmaceutical application for the dissolution and extraction of a tablet for HPLC analysis. This method showed excellent repeatability with a %rsd of 0.7 % (n=9).

It was shown that by using the CF200 Centrifuge and mVorx vortexer instrumentation, it is possible to fully automate difficult sample preparation for LC and GC applications.

Volatile organic (and inorganic) compounds are universal in chemical communication within and between virtually all living species. They are integral to carbon, nitrogen and sulphur flux through the biosphere and a significant source of atmospheric pollution and greenhouse gas emissions.

An Integrated Platform Technology for Volatile Biomarker Discovery

Changes to the pattern of individual’s volatile metabolites (volatilome) are observed in, and may be used to understand the biochemical processes associated with, metabolic disorders (including those resulting from nutrition, lifestyle choices) and disease (cancer, immune and inflammatory, infection and pathogenesis).

The Anatune Metabolomics Prep Station

Robust and reproducible sample preparation methods are fundamental to the delivery of high quality metabolomics and metabolic phenotyping datasets. Whilst the last decade has seen great improvements in instrumental precision, sensitivity and robustness; analytical throughput is limited by the twin bottlenecks of sample preparation and data processing: both are labour intensive and time-consuming.

The Use of Low Energy EI in the Workflow for the Improved Identification of Extractables

During a controlled extraction study, a range of possible identification scenarios exist; confirmed, confident, tentative and unknown. The main aim is to improve the identification threshold to a point that a toxicologist can assess if the extractables are potentially hazardous to human health.

The approach described in this poster utilises a fully automated extraction process, seamlessly integrated with the GC-MS analysis.

Development of an Automated Controlled Extraction System

The selection of solvents can be key in the development of controlled extraction studies, so the correlation between extractables and leachables can be achieved.

This poster describes full automation of the extraction workflow using the GERSTEL MultiPurpose Sampler (MPS).

Development of a Fully Automated, High Throughput Platform for Metabolic Phenotyping of Amino Acids

Amino acids are both key metabolites in primary metabolism and the structural building blocks of proteins. Robust and reproducible, routine amino acid analyses are important across a range of sectors including crop breeding, agri-food, diagnostic medicine, biomedical and epidemiological (nutrigenomic) studies, routine food testing (labelling and regulatory compliance), industrial biotechnology, synthetic biology and protein/peptide characterisation/quality control.

Extending SIFT-MS to Detection of Semi-Volatiles on Surfaces

Selected Ion Flow Tube Mass Spectrometry (SIFT-MS) is a real-time analytical technique that detects volatile organic compounds (VOCs) and certain inorganic gases to low part-per-trillion concentrations (by volume; pptv) without sample preparation or pre-concentration. These characteristics mean that SIFT-MS has traditionally been applied to real-time detection of VOCs in air.

The recent development of a swab desorber inlet has enabled SIFT-MS to be applied to detection of diverse semi-volatile organic compounds (SVOCs) swabbed from surfaces. In this study, we investigated the performance of swab-based SIFT-MS analysis of a variety of explosives, explosives taggants, drugs of abuse and their precursors, herbicides and pungent odour compounds.

Real-Time Analysis of United States EPA Method TO-14A Compounds using SIFT-MS

Selected Ion Flow Tube Mass Spectrometry (SIFT-MS) is a real-time analytical technique that offers potential for rapid screening of volatile organic compounds (VOCs) to ultra-trace levels in air.

Quantitation limits in the low part-per-trillion range (by volume; pptv) are achieved without sample preparation or pre-concentration and results compare well with those obtained at an accredited laboratory using the United States’ Environmental Protection Agency (US EPA) TO-15 Compendium Method.

This paper presents the results obtained during development of a rapid analytical method that targets compounds in the US EPA TO-14A Compendium Method.

Automated Extraction of Mycotoxins from Crop Samples using Disposable Pipette Extraction (DPX) and Analysis by LC/MS/MS

The automated clean-up of mycotoxins residues from corn sample extracts using disposable pipette extraction (DPX) is described. DPX is a solid-phase extraction (SPE) technique that is based on loosely contained sorbent inside a pipette tip fitted with a screen. This divide provides faster extraction because only minimal conditioning steps are needed which limits solvent use. A weak anion exchange sorbent (DPX-WAX) was found to provide selective extraction of 6 different mycotoxins from crop samples. Recovery of the analytes of interest averaged 84% with a relative standard deviation of 6%.

Analysis of Volatile Leachables by Automated Headspace solvent Micro-Extraction (aHSME)

The need for increased sensitivity and selectivity is a constant challenge facing many industries. One such technique that is becoming more popular amongst analytical scientists to overcome these challenges in a variety of sample types, particularly complex, dirty matrices is Headspace Solvent Micro-Extraction (HSME).

Preliminary Evaluation of ATEX-PTV-GC-QTOF-MS for the Determination of Pesticides in QuEChERS Extracts of Samples Containing High Concentrations of Chlorophyll

The GC-MS analysis of pesticides in green commodities is challenging because non-volatile components build up in the inlet liner causing gradual deterioration of chromatographic performance (tailing peak shapes and irreproducible response).

Determination of Trace (pg/L) Levels of Cypermethrin in Water using Stir Bar Sorptive Extraction

New limits for the levels of permitted pesticides in water present a challenge to the testing laboratories. For Cypermethrin, due to its high aquatic toxicity, a detection limit of 0.01 ng/L is proposed. A method is therefore required that provides a high level of enrichment, in order to avoid the need for large sample volumes using traditional liquid-liquid extraction approaches. The use of a selective GC-MS/MS method further enhances selectivity and hence sensitivity.
